AboutDNAMethyltransferases
Theadditionofmethylgroupsiscarriedoutbyafamilyofenzymes,DNAmethyltransferases(DNMTsorDNAMTases).ChromatinstructureinthevicinityofgenepromotersalsoaffectsDNAmethylationandtranscriptionalactivity.DNMT1,DNMT3A,andDNMT3BarerequiredfortheestablishmentandmaintenanceofDNAmethylationpatterns.Twoadditionalenzymes,DNMT2/TRDMT1andDNMT3L,mayalsohavemorespecializedbutrelatedfunctions.DNMT1appearstoberesponsIBLeformaintenanceofestablishedpatternsofDNAmethylation,whileDNMT3Aand3Bseemtomediateestablishmentofnewordenovo,DNAmethylationpatterns.DNMT2/TRDMT1wasshowntomethylatetRNAAspatC38andDNMT3LisfoundtobeacatalyticallyinactiveregulatoryfactorofDNAmethyltransferases,whichisessentialforthefunctionofDNMT3AandDNMT3B.DiseasedcellssuchascancercellsmaybedifferentinthatDNMT1aloneisnotresponsibleformaintainingabnormalgenehypermethylationandbothDNMT1andDNMT3Bmaycooperatewiththisfunction.
