Principle&Procedure
TheMethylamp™CoupledDNAIsolationandModificationKitcontainsallreagentsrequiredforDNAisolationandbisulfiteconversion.DNAisisolatedandchemicallydenaturedtoallowbisulfitereagenttoreactspecificallywithsingle-strandedDNA,therebydeaminatingcytosineandcreatingauracilresidue.TheuniqueDNAprotectionreagentscontainedinthemodificationbuffercanpreventthechemicalandThermophilicdegradationofDNAinthebisulfitetreatment.Thenon-toxicmodifiedDNAcapturebufferenablesDNA totightlybindtothecolumnfilter,thusDNAcleaningcanbecarriedoutonthecolumntoeffectivelyremoveresidualsodiumbisulfiteandsalts.ModifiedDNAthencanbeelutedandstablystoredat-20°Cforupto2months.
